The study investigates the current state, floristic composition, and ecological characteristics of Gentiana tianschanica populations in the Ile-Alatau region, part of the Ile-Alatau National Park, Kazakhstan.Gentiana L., an ancient boreal genus, includes 31 species in Kazakhstan, such as the endemic Gentiana dschungarica.Known for medicinal uses, especially for gastrointestinal and urogenital disorders, interest in gentians has risen for ornamental and pharmacological applications.Nine natural populations of G. tianschanica were studied in diverse ecological and phytocoenotic conditions of the Ile-Alatau mountains.Soil analyses revealed significant differences in the physicochemical properties of the species' habitats. In the Kimasar Gorge, mountain-meadow chestnut soils with a light loamy texture were observed, while the Big Almaty Gorge featured dark chestnut soils rich in humus (7.36-18.78%)and nutrients such as nitrogen (120.4-140.0mg/kg), phosphorus (58-98 mg/kg), and potassium (470-1000 mg/kg).These findings emphasize the species 'adaptation to varying altitudinal and climatic zones.Phytochemical analysis showed G. tianschanica is rich in bioactive compounds, including phenolics, bitter glycosides, flavonoids, and alkaloids.Total phenol content reached 1567.17-9008.20mg of gallic acid/100 g, with the highest levels in leaves.Flavonoids (up to 1358.25 mg catechin/100 g) and vitamin C (up to 244.7 mg/100 g) were significant.Syringic acid (1685.8mg/100 g), quercetin (317.4 mg/100 g), and chlorogenic acid (237.5 mg/100 g) highlighted antioxidant and antiinflammatory potential. The antioxidant activity, particularly in leaves, reached 530.30 mg Trolox/100 g.Anthropogenic threats, including overgrazing and habitat disturbance, have led to declining populations, necessitating conservation strategies like habitat monitoring, protective regimes, and sustainable medicinal plant use. This study underscores the ecological and phytochemical significance of G. tianschanica, which is vital for biodiversity conservation and pharmaceutical development.

This study investigates the phytochemical composition and biological activities of six solvent extracts-hexane, ethyl acetate, and hydroethanolic fractions-from the roots and aerial parts of Ajania fastigiata (C. Winkl.) Poljakov (Asteraceae), collected in the mountain regions of southeastern Kazakhstan. Phytochemical analysis revealed that the aerial ethyl acetate extract contained the highest levels of bioactive metabolites, including phenolic compounds (34.22 ± 1.98 mg GAE/g), flavonoids (2.74%), organic acids (1.91%), and coumarins (0.54%). Antioxidant potential was evaluated using DPPH and ABTS radical-scavenging assays, with values of SC50 = 133.98 ± 12.91 µg/mL (DPPH) and 209.21 ± 3.70 mg Trolox/g (ABTS). The minimum inhibitory concentration (MIC), determined by the broth microdilution method, showed strong antimicrobial activity of the aerial ethyl acetate fraction, particularly against P.aeruginosa (MIC = 9 µg/mL) and E.faecalis (MIC = 39 µg/mL). Cytotoxicity was assessed by the MTT assay on NIH-3T3, K562, and Saos cell lines. The aerial ethyl acetate fraction exhibited selective cytotoxic effects, with IC50 values of 210 µg/mL for K562 and 360 µg/mL for Saos, while showing negligible activity toward normal fibroblasts (IC50 > 2500 µg/mL). These results confirm the pharmacological potential of A.fastigiata as a promising natural source of antioxidant, antimicrobial, and anticancer agents.

Euphorbia yaroslavii Poljakov. is a rare plant species endemic to Kazakhstan whose phytochemical composition has remained largely unexplored. This study presents the first organ-specific phytochemical and GC-MS characterization of the leaves, stems, and roots of this species. Quantitative analyses indicated pronounced metabolic differentiation among plant organs. Roots were characterized by high contents of total phenolics (58.2 ± 2.5 mg GAE/g DW), coumarins (2.48%), and mineral components (total ash 13.22%), while leaves exhibited the highest tannin content (2.35%). The maximum flavonoid content was observed in roots (0.27%), whereas stems contained the highest level of ascorbic acid (0.70 mg/g DW). GC-MS profiling tentatively identified 68 compounds in leaf extracts, 65 in stem extracts, and 39 in root extracts. Root extracts were dominated by fatty acids and triterpenoid-sterol constituents, including octadecanoic acid (18.9%), hexadecanoic acid (17.5%), β-amyrin (5.2%), and lanosterol (4.9%). Overall, the results indicate organ-specific metabolic specialization in Euphorbia yaroslavii Poljakov. and provide a reliable phytochemical baseline for future bioactivity-oriented and conservation-related studies.

Atraphaxis virgata and Atraphaxis pyrifolia are xerophytic species of the Polygonaceae family that remain insufficiently characterized from pharmacognostic, phytochemical, and toxicological perspectives. This study provides an integrated evaluation of both species through anatomical authentication, sequential extraction of CO2-extracted residual biomass, GC–MS and LC–MS metabolite profiling, and acute oral toxicity assessment. Anatomical analysis revealed shared xeromorphic traits, including cuticular protection, dorsiventral mesophyll organization, structured vascular bundles, and calcium oxalate druses. It also identified species-specific differences in leaf thickness, mesophyll arrangement, vascular architecture, and druse morphology. GC–MS analysis showed distinct chemical profiles: A. virgata displayed a concentrated profile dominated by acetophenone- and benzofuran-related constituents, whereas A. pyrifolia showed a broader spectrum of carbohydrate-derived, phenolic-related, and oxygenated constituents. LC–MS analysis supported the tentative annotation of diverse polyphenolic classes, including flavonoids, phenolic acids, coumarins, and phenylpropanoid derivatives. Acute oral toxicity testing showed no mortality at doses up to 2000 mg/kg, supporting a low acute oral toxicity classification under the tested conditions. However, histological examination revealed mild to moderate dose-dependent alterations in liver and kidney tissues at higher doses. The novelty of this work lies in linking diagnostic anatomical traits, species-specific metabolite patterns, residual biomass valorization, and preliminary safety evidence within a single comparative framework. These findings provide a basis for pharmacognostic authentication, phytochemical standardization, and future bioactivity-guided evaluation of Atraphaxis species.
